چکیده

12 Most neurons have complex morphologies with long processes and dynamic, nonuniform spatial expression patterns of subcellular organelles and macromolecules. It is thought that trafficking and delivery of this subcellular cargo depends on purely local signals, rather than a global addressing system. Yet it remains unclear how such a decentralized system performs in complex morphologies. We mathematically formalize a previously proposed “sushi belt” model of microtubule transport (Doyle and Kiebler, 2011) and show how arbitrarily complex spatial distributions of cargo can be achieved by local signaling mechanisms. We reveal that this model predicts an unavoidable and physiologically critical tradeoff between speed and precision of cargo delivery that can be tested experimentally. More sophisticated variants of the sushi belt model can provide both fast and precise transport; however, these require global tuning of trafficking kinetics, and their performance is fragile to changes in the required spatial distribution of cargo. 13

Dendritic trafficking faces physiologically critical 1 speed - precision tradeoffs

12 Nervous system function requires intracellular transport of channels, receptors, mRNAs, and other cargo throughout complex neuronal morphologies. Golgi-independent secretory trafficking through recycling endosomes in neuronal dendrites and spines

Neurons face the challenge of regulating the abundance, distribution and repertoire of integral membrane proteins within their immense, architecturally complex dendritic arbors. While the endoplasmic reticulum (ER) supports dendritic translation, most dendrites lack the Golgi apparatus (GA), an essential organelle for conventional secretory trafficking. Thus, whether secretory cargo is locally ...

SPECIAL SECTION BRIEF REPORT Autism and the Extraction of Emotion From Briefly Presented Facial Expressions: Stumbling at the First Step of Empathy

Identification of other people’s emotion from quickly presented stimuli, including facial expressions, is fundamental to many social processes, including rapid mimicry and empathy. This study examined extraction of valence from brief emotional expressions in adults with autism spectrum disorder (ASD), a condition characterized by impairments in understanding and sharing of emotions. Control par...
